Precision oncology drugs often fail for reasons that are chemical as much as biological. A molecule may inhibit its target in a dish yet lose usefulness because it does not reach the brain, is limited by off-target toxicity, or cannot bind after a resistance mutation changes the pocket.

Zidesamtinib, marketed as Jideytro, was designed around those constraints. Its approval for previously treated ROS1-positive non-small cell lung cancer gives clinicians another option after earlier ROS1 tyrosine kinase inhibitors and offers a test of Nuvalent's broader design philosophy: build kinase inhibitors for selectivity, central nervous system penetration, and resistance coverage from the outset.
